The perfect combination of chemiluminescence and colloidal gold immunochromatographic test strips

Recently, Jiang Xingyu, a researcher at the National Nanoscience Center, and his research team designed a fast detection device that perfectly combines chemiluminescence with colloidal gold immunochromatographic test strips. The study was published in the journal Analytical Chemistry under the article entitled "A self-contained chemiluminescent lateral flow assay for point-of-care testing".
Chemiluminescence (CLIA) immunoassay is widely used in the diagnosis of tumor markers, infectious diseases, endocrine functions, hormones, etc. by accurate quantitative analysis of various trace active substances in human serum. With high signal-to-noise ratio, wide linear range and simple instrument, it is recognized as an advanced immunodiagnostic technology in the world, providing an important basis for clinicians to accurately diagnose diseases. However, the conventional hydrogen peroxide-luminol liquid chemiluminescence system requires separate storage of the two components and mixing at the time of use, which causes inconvenience for point of care testing (POCT). POCT is one of the fastest-growing sub-sectors in the in vitro diagnostic industry in recent years. It eliminates the complicated procedures of laboratory testing and quickly obtains test results through on-site sampling and real-time analysis. Colloidal gold immunochromatographic test paper is a very common instant detection device. It has many advantages such as simple operation, fast response, low cost and good stability, but its detection sensitivity is not high and its ability to quantitative analysis is lacking. How to combine chemiluminescence with colloidal gold test strips to make detection accurate and fast?
The researchers proposed a device that consists of three parts: a normal lateral flow assay (LFA) strip, a chemiluminescent substrate pad, and a polycarbonate (PC) scaffold. Label horseradish peroxidase (HRP) and gold nanoparticle (AuNP)-labeled antibodies on the LFA strip; for substrate matrices, using solid phase sodium borate instead of hydrogen peroxide as oxidant for the luminol system, after mixing sodium perborate with luminol, it is fixed on the glass fiber by a freeze-drying method as a solid phase chemiluminescent substrate, and the chemiluminescent substrate on the glass fiber is freeze-dried. The combination of the two can be used for highly sensitive quantitative analysis of target molecules.
The system not only achieves fast detection, but also ensures the sensitivity and accuracy of the detection. It is reported that the researchers have used it to analyze the content of folic acid in serum alpha-fetoprotein and milk powder, and obtained good results. This method opens up new ideas and methods for the application of chemiluminescence analysis in the field of POCT.
