Application of BSA|CAS:9048-46-8| in Nanotechnology
BSA is a globulin in bovine serum containing 607 amino acid residues. BSA is a commonly used PCR amplification enhancer and stabilizer, which can stabilize amplification enzymes and reduce PCR inhibitors. It is commonly used in nucleic acid detection reagents and is widely used in biochemical research, genetic engineering and medical research. Patent CN105199716B discloses a one-step method for preparing 3-mercaptopropionic acid-bovine serum albumin-gold nanocluster fluorescent material. In this method, bovine serum albumin is used as a stabilizer, and its optimal technical scheme is as follows:
2.5 mL of BSA with a concentration of 50 mg/mL was mixed with 2.5 mL of a 10 mmol/L chloroauric acid solution, and then 0.25 mL of a 1 mol/L sodium hydroxide solution and 0.25 mL of a 4 mol/L solution were added. The 3-mercaptopropionic acid solution was shaken and mixed, and reacted at 4℃ for 1 h. The reaction solution changed from light yellow to colorless to obtain 3-mercaptopropionic acid-bovine serum albumin-gold nanoclusters.
The reaction solution was dialyzed with a dialysis bag with a molecular weight cutoff of 7000 in 20 mmol/L phosphate buffer at pH 3 for 48 h, and then continued to dialyze in deionized water for 12 h to obtain purified 3-mercaptopropionic acid-bovine serum albumin-gold nanoclusters. The obtained purified 3-mercaptopropionic acid-bovine serum albumin-gold nanoclusters were colorless and transparent liquids, and had strong orange-yellow fluorescence under ultraviolet light (365 nm) irradiation.
