Product Introduction:
TRIS-HCl is an important biological buffer and can also be used as an organic synthesis intermediate. In addition, it can also be used for the growth of protein crystals under different pH conditions and the formation of intermediate fibers of nematode lamin.
TRIS-HCl Application Field
Monkeypox is a zoonosis caused by Monkeypox virus. Monkeypox virus is similar to vaccinia virus and smallpox virus. Its clinical manifestations are fever after 10-12 days of exposure and skin rash after 2-3 days of fever. The rash changes from maculopapules to small blisters and pustules, and scabs appear about 10 days later. Viruses can be transmitted from animals to humans through direct and close contact, mainly through blood, bodily fluids, skin or mucosal wounds. Secondary transmission in humans is mainly through airborne droplets.
At present, there is no universal, safe and rapid serological diagnosis method for smallpox and Monkeypox virus infection in the world, while biological differential diagnosis usually takes 3-6 days, and there are potential biological safety hazards. Due to the low sensitivity of serological diagnosis and clinical diagnosis, it is difficult to differentiate and diagnose smallpox, monkeypox, and other diseases with rash. Therefore, molecular diagnosis (PCR or fluorescence quantitative PCR) technology has been rapidly developed for rapid and specific diagnosis of viral infections. However, most existing molecular diagnostics have the drawback of low sensitivity. Therefore, CN114752711B patent developed a composition, kit, method and use for detecting and typing Monkeypox virus. TRIS-HCl is mainly used as a component of PCR buffer.
The test kit mainly contains the following components: Taq enzyme, Uracil glycosylase, Mg2+, Mn2+, dNTP, primer, probe and PCR buffer.
The common PCR buffer consists of buffer systems such as TRIS-HCl, MgCl2, KCl, and Triton X-100. Generally, the total volume in a single PCR reaction tube ranges from 20µL to 100µL.
