Indole -3- carbinol (700-06-1) through inhibiting the expression of Survivin induced apoptosis of human nasopharyngeal carcinoma cell
Objective: To explore the effect of indole -3- carbinol induced proliferation of human nasopharyngeal carcinoma CNE1 cells in vitro experimental conditionsand its effect on inducing apoptosis of CNE1 cells and the possible mechanism.
Methods: the water-soluble tetrazolium (WST-1) assay for the detection of indole -3- carbinol inhibitory effect on the proliferation of human nasopharyngeal carcinoma cell line CNE1 in vitro; method of direct observation and reflection of CNE1 cell apoptosis induced by detecting the indole -3- carbinol mitochondrial membrane potential and DAPI fluorescence staining; immunoblotting(Westernblotting) signal molecular related apoptosis survivin (survivin) protein expression.
Results: the indole -3- carbinol can dose dependently inhibited the proliferation of human nasopharyngeal carcinoma cells in vitro, and can induce cell apoptosis,and during apoptosis, inhibitor of apoptosis protein survivin expression decreased.
Conclusion: indole -3- carbinol can inhibit the proliferation of humannasopharyngeal carcinoma CNE1 cells and induce apoptosis in vitro, and the apoptosis may be related to survivin expression suppression.
