来源:亚科官网
Protease inhibitors are compounds that are ubiquitous in plants, animals and microorganisms and exert immunomodulatory, anti-inflammatory, anti-oxidative, anti-tumor, cardiovascular, and pest-resistant effects by inhibiting the activity and function of various proteases. Protease inhibitors include proteinaceous protease inhibitors and other natural small molecule protease inhibitors.
What is the protease inhibitor Cocktail
The protease inhibitor cocktail is a mixture of protease inhibitors formulated with six independent protease inhibitors (without EDTA) in an optimized ratio. It is widely used in various proteases and can be called a broad-spectrum protease inhibitor.
Action principle of protease inhibitors
By inhibiting the activity of proteases, protease inhibitors reduce the decomposition of proteins by proteases, thereby effectively increasing the rate of protein production without changing the properties of cells and tissues themselves.
The protease inhibitor cocktail contains six independent protease inhibitors (without EDTA): AEBSF, Aprotinin, Leupeptin, Bestatin, E-64, Pepstatin A. The targets of their action are serine protease, serine cysteine protease, aminopeptidase, cysteine protease and aspartic protease, etc., which act extensively on various proteases.
Use of the protease inhibitor cocktail
Protease inhibitor cocktail is suitable for protein extraction and purification of mammalian cells and tissues, Western Blot, co-immunoprecipitation (Co-IP), immunofluorescence (IF), immunohistochemistry (IHC), kinase assay (kinaseassay) ) and antibodies, enzyme diagnostic kits (Dignose Kit), etc.
During the experiment, Cocktail was pre-added to the prepared experimental system at a volume ratio of 1:100, and gently mixed uniformly.
