Methods for preparing acetylthiocholine iodide

Acetylthiocholine iodide is a substrate of acetylcholinesterase and an important diagnostic reagent raw material. The CAS.number of acetylthiocholine iodide is 1866-15-5. Acetylthiocholine iodide is often used to determine the activity of acetylcholinesterase by colorimetry. It is used together with the color developer 5,5'dithiobis(2-nitrobenzoic acid) DTNB can quickly determine the toxicity level of pesticide residues .

Sun Haoyi et al. optimized the synthesis method of acetylthiocholine iodide and developed a method with simple operation, good safety, and easy availability of raw materials, including the following steps: Firstly, the dimethylaminoethane compound is dissolved in the alkali to obtain the alkaline aqueous solution, and then the thioacetic acid compound is added to cause the thio reaction between the dimethylaminoethane compound and the dimethylaminoethyl thioacetic acid to generate dimethylaminoethyl thioacetic acid; then the dimethylaminoethyl thioacetic acid is separated and the organic solution is prepared; finally, the organic iodine is added into the organic solution to produce acetylthiocholine iodide.

The method has the advantages of simple operation, good safety, easy access to raw materials, low cost and excellent performance, which is suitable for industrial production.
