Tris(2-carbonylethyl)phosphorus hydrochloride (TCEP-HCL) is a colorless, odorless, and weakly reducing substance that is easily soluble in water,and its CAS no.is 51805-45-9. It has high stability in air and aqueous solutions, and be less affected by temperature and concentration. TCEP-HCL has mild reaction activity, is easy to dissolve, has low toxicity and is easy to handle. It is an important and versatile chemical reagent.
TCEP-HCL is a new type of reducing agent and deprotecting agent for thiolated DNA. Sulfhydryl DNA terminal sulfur atoms tend to form dimers in the solution, especially in the presence of oxygen, this dimerization greatly reduces some coupling reaction experiments (such as DNA immobilization in a biosensor) effectiveness. Adding TCEP-HCL to the DNA solution and removing it after a period of reaction can reduce the dimerization of DNA.
TCEP-HCL is also often used for the reduction of disulfide bonds in proteins, and can be used to prevent intramolecular or intermolecular disulfide bonds formed between cysteines in proteins. It is more stable than DTT in mass spectrometry applications, and has a strong reducing ability,the effect can be maintained for 2-3 weeks. It is considered to be an ideal substitute for DTT.
