DTNB has different stability under different pH values
DTNB is relatively stable in acidic solutions. When the pH value is less than 4, the absorbance at 412nm wavelength of DTNB does not change after 24h. In the two buffer systems of pH4 and pH5, the absorbance at 412nm wavelength increases slightly after 24h. As the pH of the medium increases, the disulfide bond of DTNB is gradually broken and cracked into TNBA dianions. When the pH is 9, the cracking rate will increase sharply.
DTNB has a wide range of uses
DTNB is a common color reagent for detecting sulfhydryl or thiol. It is used for colorimetric determination of sulfhydryl in biological samples. The principle is that in the presence of sulfhydryl compounds, colorless DTNB will be converted into yellow 5-mercapto-2- Nitrobenzoic acid and 5-mercapto-2-nitrobenzoic acid have a maximum absorption at 412 nm, and the absorption spectrum of DTNB does not interfere with the determination of sulfhydryl groups. In addition to being a sensitive reagent for the determination of sulfhydryl groups in protein and peptide tissues, 5,5'-dithiobis(2-nitrobenzoic acid) is also used for the colorimetric determination of uric acid and cholesterol, the determination of chlorodimethyl arsenic, and organophosphorus,monitoring of pesticide poisoning (cholinesterase determination), etc.
Precautions for using DTNB reagent
DTNB is irritating to skin and eyes, and may also cause respiratory irritation.
